Question

l'here is a great deal of interest in enzymes that can function un environmental conditions. Fukushima et al. (Extremophiles the properties of a starch-degrading amylase from Haloarcu archaebacterium) that could function over a range of p can function under extreme Extremophiles 9: 85-89, 2005) described on over a range of pH values in 4.3 M NaCl in the S-1 (an la sp. strain presence of chloroform. The results are shown in the following figure. 100 60 40 20 pH Fig. 1 Effect of pH on amylase activity in the absence (illed circle) or presence (open circle) of chloroform. Amylase was incubated for 30 min in buffers of various pH values containing 4.3 M NaCl in the absence or presence of chloroform, and activity was measured. Citrate-phosphate buffer (0.I M) was used for pH values between 4 and 6, Tris-HCI buffer (0.1 M) for pH values between 6 and 8, and glycine-NaOH buffer (0.1 M) for pH values between 8 and 10 Describe the effects of pH on the enzyme's activity. What is the pH optimum for this enzyme? What is the effect of chloroform on the enzyme's activity? Why were three different buffers used in these experiments? (4 points)

Explanation / Answer

Enzyme activity will be highly active at certain pH called optimum pH.Here the optimum pH is 6-7.

At acidic and basic pH enzyme activity is low and as pH approches to neutral range ,that is 6-7 the activity seem to rise steeply.

Chloroform act as a enzyme inhibitor, since itdecrease the activity of the enzyme.(its graph is always below )

Buffers are solutions which resist change in pH .

Citrate-Phosphate buffer maintain pH in acidic range(4-6).

Tris-HCl buffer maintain pH in neutral range(6-7).

Glycine-NaOH buffer maintain pH in basic range (8-10).
