Question

HW7. Next week you will be purifying your product using column chromatography. Review the slurry pack technique for preparing to run a column from your notebook. Place the following steps in chronological order. (1-12) Add eluent to wet sand then open and close open stopcock Tap glass as silica settles Fill column with the eluent solvent Add cotton/glasswool to lightly plug column hole (only if frit not present) Weigh silica in a beaker and make a slurry using the eluent Drain eluent until just at the top of silica then close stopcock Collect fractions Load sample Add silica slurry to column and open stopcock Add a protective sand layer Add sand until it reaches full diameter of column (only if frit not present) Allow sample to adsorb into silica by opening stopcock

Explanation / Answer

The proper chronological order for column chromatography is:

1)Add cotton /glasswool to lightly plug column hole. (so that silica doesn't escape from the column opening)

2)weigh silica in a beaker and make a slurry using the eluent.

3) add silica slurry to column and open stopcock.

4) drain eluent until just at the top of the silica then close stopcock.

5) tap glass as silica settles.

6) load sample

7) allow sample to adsorb into silica by opening stopcock.

8) add a protective sand layer. (so that column bed of the sample doesn't disturb while pouring the solvent)

9) Add sand until it reaches full diameter of column.

10) add eluent to wet sand then open and close open stopcock (this is done in order to settle the sand).

11) fill column with the eluent solvent.

12) collect fractions.
