A-1: Enzyme binds to the substrate to form the activalted complex. Without the substrate no enzyme activity will be found.

A-2. When you boil the enzyme, the enzyme molecule is denatured and t loses its active sites. The enzyme's activity is regulated by the active sites in it. As the boiled enzyme has less active sites than the normal enzyme, test tube 5 would have lower activity than 3.

As the temperature of the system increases, the reaction becomes faster and more of the activated complex (between substrate and enzyme ) forms and the recorded absorbance also increases.
