Question
The balance between protein biogenesis and degradation is a complex process in the cell that involves structural and regulatory networking (see figure below. Proteome balance Protein production Protein depletion Proteome imbalance productionProtein (a) List 4 structural or regulatory proteins that are important for protein production and 4 that are Protein depletion important for protein degradation. (b) What happens to these elements when there is an imbalance in the proteome? And what are some downstream implications of this imbalance?Explanation / Answer
A)Structural and regulatory Proteins that are important for protein production are:
RNAP2(RNA POLYMERISE 2) Protein : it catalysed the transcription of DNA to synthesis precursor mRNA.
TAT: it associate with RNAP2.
Histone:it is help in post translational modification that ease of interaction of DNA and domain of protein.it undergo modification leads to so called histone code which acts in different processes like gene repairs etc.
CAP(Catabolite activator protein) it attach with histone unit and helps to protein synthesis.
Protein leads to degradation of proteome
Ubiquintin: ubiquintination leads to degradation by preventing protein interaction.
HSP27 :It catalysed the degradation by binding to ubiquinated protein.
CyclinB1 : it helps in cell division processes after degradation of protein by ubiquintination pathway.
CDKI :It is an inhibitor which linked with CyclinB1.
B)The imbalance in proteome leads to switch down some regulatory enzymatic functions.
This cause ageing means progressive decrease of proteostatics. And this imbalance leads to widespread protein aggregation with abundant Proteins that exceed solubility contributions in cell.
However,an important functioning of these system is to prevent accumulating potentially toxics misfiled and aggregated protein species and this compromises the defense mechanism against reacting species.
