Question
If you mix 100 mu L of a sample with 200 mu L of buffer, what fold dilution is this? If you take 100 mu L of the dilution from a and add a second 200 mu L of buffer, now what is the fold dilution? If you perform this same dilution approach a total of 4 times, what is the final fold dilution? If your final dilution from part c has a concentration of 0.00500 M. what was the concentration before these serial dilutions? (Remember that the fold dilution is an exact number and thus significant figures do not apply)Explanation / Answer
3.
a. Mixing 100 uL with 200 uL gives a total volume of 300 uL
so dilution is 3 fold.
b. If 100 uL of solution a is diluted with 200 uL of buffer
so this dilution 6 fold overall
c. If we take 100 uL of b and dilute with 200 uL buffer
so this dilution 9 fold overall
If we take 100 uL of c and dilute with 200 uL buffer
so this dilution 12 fold overall
d. If solution concentration after 4 dilutions is 0.005 M
Concentration initislly of solution = 0.005 x 12 = 0.060 M
