1) In what ways does the [absorptionspectra] for both bound/unbound dyes different and why is this difference important for using the dye to determine [protein concentration]? Once determining the [proteinconcentration] of an unknown, would you expect to get the same answer no matter what [unknowndilution] was used, why or why not? Why could 2 [amino acids] be different at numerous positions in their [amino-acid sequence] and still be very similar in [structure/function]?

when a dye binds to a protein the [absortionspectra] will only read the dye that is bound to the protein. so in essence you are measuring the protein concentration by measuring how much dye bonded to the protein. This concentration would only be specific to that dilution, if you used an unknown dilution it would have a different Concentration because it has a different amount of protein. An amino acid sequence can be several hundreds of amino acids in a certain order. simply changing the location of 1 or 2 amino acids could change the function or structure to a certain degree. But alot of the amino acids work collectively together to provide a certain function., missing or replacing one single amino acid may have no effect to the amino acid sequence due simply to the size of the chain.
