If enzyme 3 was non-functional than there would be accumulation of substrate C and no substrate D would be formed.
however it would not lead to the cessation of entire metabolic pathway as long as the initial substrate A is available because other enzymes are still functional. also,the cell might survive if it is able to obtain the essential amino acid from the its surroundings.
