1) Each of the 15 individual amino acids you worked with in lab is soluble to some degree inwater. They had to be if they were to solubilize in the water attached to the chromatography paper. Look at the general structure of an amino acid - what specific parts of the general structure of an amino acid (functional groups) that are common to all 15 amino acids make each of them soluble in water? Explain why these specific functional groups are soluble in water – be specific. 15 Amino Acids: Isoleucine, Leucine, Phenylalanine, Valine, Tyrosine, Tryptophan, Alanine, Threonine, Serine, Arginine, Glycine, Glutamine, Glutamic Acid , Aspartic Acid, Lysine

2) What is the relationship between relative mobility (Rf) and hydrophobicity? Is there a direct relationship (as one gets higher the other gets higher) or is there an inverse relationship (as one gets higher the other gets lower)?

Explanation / Answer

1) All the fifteen amino acids which are soluble in water form zwitterions. These zwitterions are capable of forming polar covalent bonds with the oxygen atom of water. This property makes these amino acids to be soluble in water.

2) The filter paper strips are made of hydrophilic substances like cellulose. So, hydrophobic molecules move a greater distance when compared with hydrophilic substances. So, there is a direct relationship between relative mobility and hydrophobicity.
