You will receive two samples related to the scenario. You will need to review all of the reading material prepare for lab, Tutorial videos, Background, protocols, instrument manuals, package inserts, etc.). The concept map (PCR and Sequencing of mtDNA) should be used to help guide you through the first project. A scientific notebook is required. The notebook should include a title that clearly describes t Purpose and background information showing why this knowledge is of interest and any literature that is relevant to the project. A testable hypothesis should be clearly stated, not a prediction. Appropriate materials and methods section should be clear enough so that anyone could read and repeat your experiment and have similar results. Data and results section should be in the correct format (table, graph) with no mistakes and shows the relationships between the data without the reader referring to the text. The narration of the results should be very clear and relevant to the hypothesis. Statistical analysis is appropriate, correct and clearly explained. Conclusions should be clearly formed from the stated hypothesis and collected data. Primary literature should be extensively used to make connections between the project and other research in the field. The implications or application of the research should be clearly stated. The notebook should be easily understood due to the excellent grammar, word usage, and organization. You will also be giving a presentation on your work and will be graded on your participation in the group. purpose : road kill creatures are the animals killed by vehicles.it is very important to be aware about their source, what made them come to the roads from their natural habitats.also about their biological structure and changes( if any).to find out out about their extinction level and find out ways to protect them .road kill creatures can be used for several research work as well. they can also be useful for for formation of useful substances for drugs , isolation of certain bacterias etc

3)the chromosomal DNA forms an insoluble clot and denatured protein and cellular debris.
4)the clot is pelleted by centrifugation and plasmid DNA soluble is received as supernatant
5)boiling will FACILITATE THE isolaion ON plasmid DNA IN UNDERGRADED FORM.
