Percentage viability = (OD of sample - OD of blank / OD of untreated - OD of blank ) x 100

Blank is the well containing media and OD media is the absorbance of blank solution

For OD sample take the average of the 3 triplicates. For example for 1 : 0.245 + 0.257 + 0.299 / 3 = 0.267 is the OD

OD untreated is the absorbance of untreated cells. In your case OD cell is the absorbance of untreated cells

(0.267- 0.079 / 0.351 -0.079) x 100

= 69.1 %

Average OD sample OD media (blank) OD cells (untreated) percentage viability 1 0.267 0.079 0.351

(0.267- 0.079 / 0.351 -0.079) x 100

= 69.1 %
