In her studies of yeast genetics, Janet isolated 5 adenine auxotroph mutants: adel, ade2, ade3, ade4, ade5. She did some crosses to get each one in mating type a and . Then she mated them in all pairwise combinations and tested the resulting diploids for their ability to grow on medium without adenine. Results are shown below, where "yes" means that the diploid can grow without adenine and "no" means it cannot. adel ade2 ade3 ade4 ade5 4 Molecular Genetics 501 ade1 no no yes yes yes ade2 ade3 ade4 ade5 Chapter 11 Practice Problems noyes yes yes no yes no no yes no (a) How many complementation groups did she find, and what mutants are in each group? (b) How many different genes are represented in her collection of five mutants? (c) Are adel and ade2 alleles? Are adel and ade3 alleles? 1126 AMExplanation / Answer
(a) Complementation means once you are deleting a gene from the organism to create a mutant followed by supplying a vector containing the same gene into the mutant, which behaves as wild. So Janet found 8 complementation groups.
The mutants in each group are (ade1,ade3), (ade1, ade4), (ade1,ade5), (ade2,ade3), (ade2,ade4), (ade2,ade5), (ade3,ade4) and (ade4,ade5).
(b) There are two different genes represented in her collection of five mutants.
Its because, all crosses are able to grow in adenine deficient media except the self mated groups. while cross between ade2 & ade1 and cross between ade5 & ade3 are not able to produce the offspring which can grow in adenine deficient media.
(c) ade1 and ade2 can be alleles. because the absence of both in an organism leads to no growth in adenine-defficient media. But ade2 and ade3 cannot be alleles, as their absence still leads to growth of the organism in deficient media.
