Ans1) nutrient agar is used ina medium because for the purpose of cultivation of wide range of microbes which supports the growth of nonfastidious organisms. It also supports the growth of certain bacteria which are used for antibiotic senstivity , for the purification of culture. it contains certain nutrients which are necessary for the gwowth of bacteria .

Ans2) For the formation of bacterial colony, bacteria are being spread onto the surface of petri dish which contains certain amount of agar which contains nutrients necessary for the growth of culture .the bacterias make use of this by consuming the agar nutrients. Then the bacteria begin to grow and multiply . Then with due course of time bacterias generates to thousands to millions . Then the pile of bacterial cells develop from one single cell and is called bacterial colony.

Ans 3) petri plates are inverted during incubation so that they can reduce the risk of airborne particles contamination onto the culture. Petri plates containing culture are also having risk of contamination through water condensation , so inverting the petri dish also helps from water contamination.

Ans 4) Here aseptic means making the culture free from any contamination that can be caused by the harmful bacteria , viruses or other microrganisms that can be through air , water ,touching the culture with naked hands etc . So aseptic helps in making the culture or any experiment free from microrganisms.
