A. Where might your Gene of Interest be inserted?
B. Apart from your Gene of Interest, what other genes/gene sections might have to be added?
Using the Ti Plasmid to Create GMO Plants The Ti Plasmid, gene guns, electroporation and microinjection can be used to create genetically modified crops (GMCs) which are healthier, resistant to insects, herbicide tolerant or produce more fruit. Some examples are: FlavrSavr tomato (1994) longer shelf life, slower ripening Tobacco plant resistant to herbicide bromoxynil (1994) BT Potato (1995) produces pesticide Cotton plant (1995) resistant to bromoxynil Soybeans (1995) resistant to glyphosphate Golden Rice (2000) vitamin-A enriched rice Agrobacterium tumefaciens use their Ti Plasmids to insert genes into plant cells in order to create a suitable environment for them. Genes on the Ti Plasmid create a 'crown gall' in the plant. The bacteria bind to the plant cell and inserts the T-DNA into a random site of the plant genome.Explanation / Answer
A) The desired DNA./gene of interest can be inserted into the T-DNA region of the Ti plasmid .
The T-region normally ranges from 10-30kilo base pairs
b) Apart from gene of interest other regions which are required includes
MCS(MULTIPLE CLONING SITE) having unique restriction endonuclease sites for cloning the target gene.
RIGHT BORDER SEQUENCE OF T-DNA -they are required by the endonuclease as a target.
ORI for replication of cloned target gene.
SELECTABLE MARKER for selection of recombinant/transformed cells.
Virulence genes- required for transfer and integration
