Question

Please read and answer the question. Please show work and try not to write in cursive. Ignore question 4 and 5 at the top.

You work in a lab that is not restricted by money, NIH or ethical concerns. You have a lot of time on your hands, so you decide to create an interspecific protoplast cell culture between a man and a spider. You want to study the resulting hybrid to determine if a superhero could really exist.Using what you have learned about protoplast fusion, come up with your protocol.(Include: specific cells used, protoplast method, hybrid selection, culturing concerns, future plans).

4. Apart from your Gene of Interest, what other genes/gene sections might have to be added? 5. How will you test for a successful cell modification? Using Protoplast Fusion to Create "Spiderman" Protoplast Cultures fuse together two cells that have no cell wall. Scientists use various mechanical (micropipette), electrical (electrofusion) or chemical (polyethylene glycol) means to encourage the cells to form a hybrid. Because these methods are random and uncontrolled, the protoplasts rarely form a true hybrid. However, if (when) a hybrid is formed, it may regenerate a cell wall, grow, and divide. This hybrid culture can expand using special media. This technique is used mostly for plants, fungi (yeast) and plant:fungi hybrids. It has been used to improve the production of cephalosporin C and increase plant resistance to certain viruses.

Explanation / Answer

Protocol photo protoplast fusion of spider cells and human cells by using protoplast fusion method

1) Take a cell from human organism and a spider cell s Since both do not have cell wall therefore diffusion is easier

2) Fuse the cells using electric shock (electrofusion )or chemical fusion to join the cells together or nuclei together .The resulting nucleus fused together is known as heterokaryon.

3) The cells are further fused with the help of hormones .

4)The hybrid cells are then grown in culture medium. They are tested for web formation.

All those cells that can spin webs are selected as recombinants while the rest are eliminated.

5)Since these are human cells proper care must be taken about the temperature of culture, the nutrient supply etc.

6)The embryo at a stage of 8 cells can then be transferred into a womb for proper gestation.

Future plans-Many specific characters of animals can be combined with human species to produce mutant human species.
