Describe the applications and limitations of albumin, enzymes, LISS, and antiglobulin in technique in crossmatching? What antibodies are expected in each of these enhancers?Explanation / Answer
Agglutination test
In Agglutination test donor's RBC being suspended in saline and tested against antibodies at room temperature
IgM antibody used
Albumin or Enzyme agglutination test
Best method for detecting Rh antibodies.
Rapid method
sensitive method for Kidd antibodies if an AHG test is done
Albumin or Enzyme Used to enhance agglutination
Basically IgG antibody used.
Disadvantage is Enzymes enhance some cold, clinically insignificant antibodies that we do not want to enhance.
Low ionic strength saline test (LISS)
Advantage
This test is Used to provide a rapid and sensitive method for detecting most blood group antibodies.
Decreased incubation time of 10-15 min.
Disadvantage
Non-specific binding of complement if ionic strength is too low.
Antiglobulin test
Antiglobin test Used to detect incomplete IgG and IgM antibodies.
As example Kidd and Duffy antibodies.
