Question

Dont worry about the 2nd one just answer the 1st one.

Please help answer these questions. WIll give great reviews. GIve explanations please.. 1. You are evaluating a gene of interest using Northern (RNA) and Western (protein) blotting. You wish to evaluate the impact of specific miRNAs and siRNAs on the regulation of your gene. You have a specific miRNA and a specific siRNA that should target your gene. You treat different samples of cells with each RNA, and then evaluate the material from these treated cells on blots, in comparison to your control treated cells. Assuming each RNA does regulate your gene, indicate the expected result on the gels, following treatment.

2. The following diagram illustrates a step in the process of translation. Add the following elements to the diagram.
a. 5’ and 3’ ends of the mRNA
b. A, P, and E sites
c. Start codon
d. Stop codon
e. amino and carboxyl ends of the newly synthesized polypeptide chain
f. Approximate location of the next peptide bond that will be formed
g. Place on the ribosome where release factor 1 will bind

Explanation / Answer

1.siRNAs are double stranded RNAs that regulate gene expression by binding to complemenatry sequences of mRNA an degrade it.miRNAs also regulate gene expression as they bind to 3' untranslated regions of specific targetted mRNA and degrade them. So,following can be concluded:

1st gel : Northern blot

Northern blot helps in detection of RNA /isolated mRNA in a sample.miRNA and siRNA both bind to mRNA and degrade it.As a result of which no bands are seen in the miRNA and siRNA lanes. As controls are not subjected to any regulation, we can see a band that corresponds to mRNA in the control lane.

2nd gel: Western blot

Western blot helps in detection of specific protein.Since there are no bands in the miRNA and siRNA ,we can say that mRNA subjected to the treatments have undergone degradation as a result of which no protein was synthesised. We can see a band in control which confirms the presence of protein that was synthesized from mRNA not subjected to any regulations.
