Help with this problem please
Chinese hamster ovary (CHO) cells are often used to produce therapeutic proteins in a fed-batch reactor. The initial culture volume is 45L. A feed stream containing essential nutrient is continuously feeding the reactor at 4L/h. The initial nutrient concentration is 250g/L, and the initial cell concentration is 20g/L. (mu max = 0.2, K_s = 0.5 g/L, Y_x/s = 0.4) What is the culture volume at t = 5 hour. What is the concentration of the nutrient at t = 5 hour. What is the total amount of cells at t = 5 hour.Explanation / Answer
This is the example of variable volume fed-batch reactor. Hence the culture volume (a) can be calculated by using formula:
dV/ dt = F (Where F = feed rate [volume/time], dV = change in volume, dt = change in time.)
dV/ 5 = 4 (since change in time 5 hr and feed rate is 4L/hr
dV = 20
Therefor the culture volume after 5 hr will be 45+20 = 65L (since the initial volume is 45L and change in volume after 5 hr is 20.)
b. Concentration of substrate can be calculated by the formula: ds/ dt = F. (s0 - s)/ V - u.x/ Yx/s . Where ds = change in concentration, s0 = substrate concentration in the feed, u = specific growth rate, x is the biomass concentration.
c. total amount of cells can be calculated by the formula = dP/dt = qp . x -–P . F / V
