Question

(a) The Profiler Plus Amplification Kit can analyze 9 STRs plus a sex chromosomemarker in one PCR reaction, using a variety of PCR primers. The products which containequivalent numbers of repeats in many cases are able to be distinguished on theelectropherogram. How does the assay determine which locus is which when the reactionproduct is evaluated using capillary gel electrophoresis? There are 2 ways, describe thembriefly, specifically discussing on the primers that are used. Hint, the sizes of DNA fragmentsdetected run horizontally on the graph.

(b) Address the question ending Case 2 above. If the three daughters had three different
paternal alleles, what would be your conclusion?

Explanation / Answer

1. ANS: The two ways are:

Length: The length of the STR will differ from loci to loci.

DNA sequence: The exact sequence of the DNA fragment will be complementary to the primer sequence. So, the sequences of the primers will also change according to the sequence in DNA.

2. ANS: If three daughters had 3 different paternal alleles, we assume that each daughter has one allele from her father which is absent in the other two daughters. Since there are 13 STR loci in the DNA, there will be more than one match between the daughters. This would prove that they are related to each other and to the father.
