Question

Genotyping exconjugants on supplemented media To make a rough map of the lya2 gy3, and phe f loci, you perform an interrupted conjugation experiment between the following two strains: Hfr lyae^+ gly3^+ phe f^+ Amp 1^+ times F lye2^- gly3^- phe2^- Amp^R You interrupt conjugation after 1 min. 2 min, and 3 min and plate exconjugants on minimal medium containing ampicilin and various combinations of lysine, glycine, and phenylalanine. You then score the resulting plates for the presence of colonies. Based on the results shown below, what are the possible genotypes of the colonies that grew on each medium? Answer the question by dragging one pink label to each pink target, one white label to each white target, and one blue label to each blue target. Labels may be used once, more than once, or not at all.

Explanation / Answer

For the first question, the genotype wil be like this-- in the pink box wil be lys2+, whereas in white box put phe1- because in the given condition phenylalanine is absent and in the blue box it will be gly3+. for the second condition, put lys2-, in the white box put phe1+ whereas in blue box put gly3+. For the last condition, in the pink box put lys2+, in the white box put phe1+ and in the blue box put gly3-.

In the conjugation process, the positive alleles will be dominating over the negative alleles. So in the given conditions the absence of the alleles like phenylalanine or lysine or glycine, you can put the negative alleles whereas present of the alleles can be shown by putting the positive alleles. Coming to the conjugates Hfr & F, you will get the Hfr cell only along with the recombined with F gene.
