If we add equal concentrations of ConA and glucose to a dilute suspension of citrated red blood cells in ConA buffer, we will not expect to see the hemagglutination reaction.ConA has high affinity for glucose residues.Con A having high affinity for glucose,the free glucose will compete with the the receptors on the Con A to bind to them thus preventing binding of Con A to glucose residues on the red blood cells.Also, if we added equal amounts of ConA and galactose to a dilute suspension of citrated red blood cells in ConA buffer,we will see the heagglutination reaction to occur.This is because the free receptors on ConA will not bind to galactose but will bind to glucose residues present on the red blood cells resulting in agglutination.
