Question

?-Lactam antibiotics only work on growing cells. Therefore any mechanism that stops the cell’s growth rate increases the resistance to Amdinocillin. Based on the information above and reading the abstract what is one reason Amdinocillin resistance so rare?

BIOL 371 Microbiology Figure 1. Mutated genes present in twelve amdinocillin resistant clinical isolates. The far left column lists the mutated genes and the top row describes the strain name for each clinical isolate. Gray shaded boxes indicate the individual genes mutated for each clinical isolate glix alas FIG 1 Putative Amd resistance-encoding genes i clinical strains. Genes marked by gray shading have a mutation that is not present in any of the reference genomes. Specific mutations are listed in Table 4

Explanation / Answer

The Amdinocillin resistant mutants grow slowly in laboratory conditions therefore, it is difficult for the mutants to become fixed in the bladder in clinical settings because of frequent repeated micturition. In other words, a high growth rate of the pathogen is required in the clinical environment, if it has to counter its constant expulsion from the urinary tract. From this idea and the information provided, we can say that cysB mutatants found in clinical isolates showed a relatively less growth reduction whereas the resistant mutants found in the laboratory selections showed severe growth rate reduction. Hence, amdinocillin resistance in clinical isolates is so rare.
