Another issue that created problems getting the pathway to run in E.coli is that the PAL reaction is reversible. In fact, the PAL reaction used industrially to make phenylalanine. In order to fix this problem, Bang, et al. looked into non-plant PAL. It is known that the bacterial version of this enzyme is controlled differently and cinnamic acid, in bacteria, is used for another purpose. They selected the PAL enzyme from Strepiomyces maritimus. They did the kinetics of both the forward and reverse reaction. Note that the reverse reaction has two substrates, both of which were assayed below. Figure 5.
