Question

To control the Lysogenic and Lytic growth of lambda phage, the lambda repressor and Cro play important roles in gene regulation for the Lysogenic and Lytic growth. To underhand the DNA binding mechanisms of these factors, you measured the complex formation between the transcription factor (X repressor or Cro) and their target DNA by using the DNA gel-shift assay and plot DNA hound (%) as a function of proteins (lambda repressor or Cro). Which one of graphs in below should you expect to obtain for each DNA binding protein? lambda repressor. A, Cro: B lambda repressor A, Cro C lambda repressor C, Cro: A lambda repressor C. Cro: B lambda repressor C, Cro: D

Explanation / Answer

b)Controller of Repressor's Operator (Cro) it does not kill the host. It is a transcription factor, it has been repressesed by cI and alsoother pro-lytic genes which are expressed from the PLpromote. Therefore forming a positive feedback loop with Cro some times it is called as double negative feedback loop but it may not correct.

During self regulation of of cI repressor levels when it is cI repressor binds to OR1 both t transcription and Cro protentiates are repressed.when cI binds to OR2,cI repressor transcription will be stimulated. When the cI repressors are high in level then the third site OR3 will be bounded by cI repressor. So overlaps the binding overlapsthe cI promotor further production of cI repressor will be blocked till its drop the levels and again the OR3 site is free.
