Question 2) You are simultaneously expressing three different proteins, A, B, and C, in the same cells – that of the bacterium E. coli. All three proteins contain a His-His-His-His-His-His peptide sequence covalently linked via a peptide bond to the native protein sequence. The polypeptides of all three proteins are ~630 amino acids long and have a pI of ~6.5. Protein A has a sequence Glu-Asp-Asp-Tyr –Asp-Gln-Gly sequence between the His-His-His-His-His-His and the native protein sequence. Protein B is known to dimerize (i.e. has a quaternary structure composed of 2 subunits), while proteins A and C are known to lack any quaternary structure. To start purifying these proteins away from all other proteins normally expressed in E. coli, you first lyse the cells and remove the insoluble cell debris. Based on what we learnt in class: A.
