You are trying to determine when and where a newly discovered Drosophila
gene is expressed during embryogenesis. You decide to use two approaches:
1) Perform in situ hybridization
and
2) Create a reporter construct in which you take a 2,000 bp-long region upstream
of the gene’s start codon and fuse it upstream of the GFP gene; introduce this
construct as a transgene into the Drosophila genome, and observe GFP
expression.
When the experiments are finished, you find that the results are not exactly
identical in both cases.
1) What is the likely reason for the discrepancy between the results produced by
two experiments?
2) Which of the two techniques is more likely to reveal the “true” pattern of gene
expression and why?

Q1:The most likely reason for the discrepency between the results produced by two experiments is a genetic phenomeno called as Position effect.As per this effect a gene in natural locus produces a default product but when the locus of the gene changes, the function of the gene changs as well.In case of our experiment where we employ In Situ hybridisation ,the natural position of the gene concerned is not changed and hence the desired product is obtained.Whereas, when a gene construct is used, it is likely that it may get incorporated at a different place in the genome and as such a different product will be obtained.
Q2.The Insitu hybridisation is more likely toreveal the true patteren of the gene expression as the locus of the gene is not changed.
