Question

2. You are examining the behavior of two centrosomal proteins Centrin and Cep192 by FRAP analysis. Both proteins are expressed as GFP-tagged forms and localize to the centrosome (the centrosome is indicated by red the arrow, the plasma membrane is indicated by the dashed line). p.b is before bleaching, the subsequent images show time points during recovery from photobleaching (time is indicated in minutes and the time course shows the fluorescent signal at the centrosome over time, always focusing on the centrosome). Which of the following conclusions can you draw from this experiment? The legend to this figure as described in the research paper by Zhu et al., 2008. Current Biology 18, 136-141) FRAP analysis of HeLa cells expressing GFP-tagged murine Centrin-2 or murine Cep192. Images were taken before (p.b.) and immediately after photobleaching (0) and every 2 min afterwards. Insets show 6-fold magnifications of the centrosomal region. The scale bar represents 10 m. (1 point) Centrin-GFP Cep192-GFP OBoth, Centrin and Cep192 are stably associated with the centrosome and do not exchange with cytosolic subunits. OCentrin-GFP displays a more dynamic behavior than Cep192 O Cep192-GFP is more dynamic than Centrin O The dynamic behavior of Cep192 and centrin is similar O The dynamic behavior of Cep192 depends on the cell cycle stage 3. What do you expect to see if you fixed the cells from question 2 (after the FRAP experiment has been performed) and performed immunofluorescence microscopy staining, using specific primary antibodies to centrin and Cep192, followed by staining with appropriete secondary antibodies? (1 point) O Centrin and Cep192 will be absent from the centrosome OOnly centrin will be absent from the centrosome, but Cep192 will still be present at the centrosome O Centrin and Cep192 will both be present at the centrosome OCep192, but not centrin, will display a high level of motility at the centrosome

Explanation / Answer

ANSWER: In my opinion also I guess for both Centrin and Cep192 are needed to be present at the centrosome. As in immunofluorescence microscopy localisation of specific antibodies is done which will be present in both the proteins where centrin may show a little reduce level of mobility but will not be absent on a whole.
