Question
Using Sanger dideoxy sequencing, you determine the DNA sequence of the mitochondrial NADH dehydrogenase subunit 2 (ND2) gene from unaffected and LHON affected individuals. The sequence data is shown in Figure 1 below. Which of the following correctly represents the nucleotide sequence differences in the ND2 gene between LHON and unaffected individuals?
Hi! I'm having a hard time trying to figure out how to solve a question like this? What sort of thinking should I have?
Thanks!

Answer - E
The unaffected patient sequence they given it is Adenine ans also the peak in green colour in unaffected patient so it is Adenine. Then, in Lhon patient the peak shown in black color. In the sequence T-Red color and C-Blue color. There is no black coolr in the given sequence so it might be Guanine
