How to do problem 1? Diluted Standard 100 UM Vol. of Standard 9.45 ul 10 mM 645 ul of 100 uM 506.25 of 80 uM Vol. of Diluent 935.55 ul 20 uM 262.5 ul 150 ul of 10 HM Problems 1. You are performing an assay to measure the nitric oxide concentration of a serum sampl perform the assay, you need a set of standards of known nitric oxide concentration. How would prepare nitric oxide standards of 5, 10, 15, 25, 30 and 50 g/ml from a 1 mg/ml nitric oxide stock by serial dilution? [you need at least 200 1 of each for the assay] e. To you ng/mL You are nerforming an assay to measure the protein concentration of a serum n fntondards of known proteinExplanation / Answer
1 micro [] = 0.001 milli [m]
1g/ml = 0.001 mg/ml
You can use this equation V1C1=V2C2
V1 = the volume of stock you will start with. This is your unknown.
C1 = 1 mg/ ml in the stock solution
V2 = total volume needed at the new concentration = 200 ul = 0.2 ml
C2 = the new concentration = 1g/ml
V1 = (V2 x C2) / C1
V1 =(.2 ml * 1g/ml)/1mg/ml
V1 = (0.2ml * 0.001 mg/ml) / 1 mg/ml
= 0.0002 ml = 0.02 ul
so take 0.02 ul of the stock solution and make the volume up to 200 ul
for the first dilution, 5 ug/ml take 0.1 ul and make up the volume
for 10- take 0.2ul
for 15 - take 0.3 ul
and so on
note - I have solved this question by taking the printed value not the one you have written with pen
