We know that enzymes are proteins and proteins are only functional in their three dimensional configuration. Lactase is also an enzyme. So,any change in the aminoacid would bring a change in the three dimensional shape of lactase that would effect its enzymatic function. A change in single aminoacid would also alter the three dimensional structure of the different versions of the lactase and this would also effect its enzymatic activity. We can go for an enzyme assay to test for the enzymatic activity of each of the different versions of lactase to find out in which aminoacid version ,the enzymatic activity is least which means that version of lactase has significant alteration in its three dimensional shape. The versions that alter the three dimensional shape of the protein would show none/least enzymatic activity oe we can say no enzyme activity means significant change in three dimensional shape of lactase.
