Lab #4: Competitive vs. Non-Competitive Enzyme inhibition A study in catechol oxidase activity Question: is PTU a competitive or non-competitive inhibitor of catechol oxid Use the data from Procedure (minutes) vs. Absorbance axis (dependent variable) added to the top of the graph. Use 3 different colors for the data from tubes 1, 2, and 3 an key. Once you have completed the graph, answer the following questions. ase? #2 (Question#3 on your lab question sheet) to construct a graph of Time at 540 nm. Make sure your scale on the X axis (independent variable) and Y are consistent, that axes are labeled properly, and that a descriptive title is 1. How does a competitive inhibitor differ from a non-competitive inhibitor? 2. Based on your data, is PTU a competitive or non-competitive inhibitor? 3, Explain, using specifics from your data, how you arrived at your conclusion in question#2.

Explanation / Answer

competative inhibitor-it binds to the active site of the enzyme to prevent the substrate binding such that reaction cannot be catalysrd and thus it slows down the rxn.it competatively binds to the active site with the substrate .the str ucture of inhibition molecule is similar to that of substrate.it does not alter the structure of enzyme.the rate can be reversed by increasing the substrate concentration.ex-sulpha drugs given to bacteria compete with para amino benzoic acid and folic acid synthesis is inhibited.

while non-comp inhibitor act at a site other than active site on the enzyme molecule so they dont compete but change the shape of an enzyme once they bind to it,such that the substrate cannot bind to the active site.the shape of enzyme changed parmanently and substrate will not bind to the active site no matter how much it is present.the structure of inhibitor is entirely different .the rxn goes on decreasing as more inhibitors contact the enzyme till saturation is reached.ex-cyanide combines with the prosthetic group of cytochromo oxidase and inhibits the electron transport chain.

2)PTU is a competative inhibitor of the enzymatic oxidation of DOPA by phenoloxidase.phenoloxidase is used for melanization catalyzing the oxidation of phenols.it is used as inhibitor of phenoloxidase.

3)data not given
