Question

BICL3SO CELL BIOLOGY PROBLEM SET 1 (50pts) Spring 2018 Name FSU ID 5. (9pts) You are studying a protein with molecular mass of 200kDa. You have constructed the following cell lines: a wild-type strain, a mutant that overexpresses this protein, and a truncation mutant that expresses a shorter sequence of this protein MW Sample 1 Lane 3 Lane 4 Lane 5 PAGE Gel 400 kDa 300 200 100 50 d contains a cell homogenate from the wid-lyne strain and you run this sample on an SDS-PAGE gel. Circle the band that represents the wild-ype protein you are studying B. In Lane 3, draw on the gel diagram what you would expect the bands would look like for a sample containing cell homogenate from the overexpression strain. (You can assume the total protein concentration is similar to Sample 1.) C. In Lane 4, draw on the gel diagram what you would expect the bands would look like fora sample containing cell homogenate from the wild-type strain, and the total amount of protein is double that in Sample 1 (ie twice the total protein concentration) D. In Lane 5, draw on the gel diagram what you would expect the bands would look like for a sample containing cell homogenate from the truncation mutant strain. (You can assume the total protein concentration is similar to Sample 1.) E. Using only the protein bands on the gel, how can you identity which band in Lane 5 coresponds to the truncated protein? he tmumcated Gena will appear 'n locater, one Gel kotner, 200 eba eut is impassible to dctermie the czact tocation oecouse amount the Gene was €nortened is dcternuine ess tnan 200uDa

Explanation / Answer

A. yes your answer is right but the band first which is present on 400 kilo Dalton size in sample first lane is also similar to wild type lane. Include both bands ( 400kd band and 200kd band)in your answer because both of bands have similarity with wildtype band.

All answers are good and exactly correct.

Well tried.
