Question

Pre-laboratory 1. In what sort of environments would you expect to find spore-forming bacteria? 2. Why is it important to determine the location of the endospore within the bacterial cell? 3. List the benefits of endospore formation for bacterial survival. 4. Can you relate endospore staining to endospore survival in hospital or other environments? 5. Explain bacterial sporulation. 6. What would the result of an endospore stain of a smear of a Clostridium species be if: a- The malachite green was not heated? b-The counterstain was omitted? c -The malachite green was not effectively rinsed? d-The final rinse after the counterstain was excessive? f- The culture of bacteria stained was only 12 hours old?

Explanation / Answer

1. All bacteria remains in their 'vegitative forms ' , if the environmental conditions are favourable for there normal metabolic activity.

The bacteria which produce a spore to service under adverse environmental conditions in a spore forming bacteria. Mostly the following three rod shaped bacteria can produce spore under adverse environmental conditions .

A) aerobic rod shaped bacteria ----> 1.Bacillus spp.

B)anerobic rod shaped bacteria ----> 2.Clostridium app.

3.Desulfotomaculum app.

2. The position of endospores differs among bacterial species and is useful in identification . The main types within the cell are terminal , sub terminal and centrally placed endospores .

3. Endospores are resistant to the most agent that would normally kill the vegetative cells the formed from.

Endospores of certain types of bacteria , such as geobacillus stearothermophilus , are used as probs to verify the auto clave the item has been rendered sterile.

Bacterial endospores are resistant to antibiotics, most disinfectants , and physical agent such as radiation, boiling, and drying.

4.In hospitals , endospores have delicate invasive instruments (eg., Video endoscopes) are killed by low temperature and non corrosive , non toxic, plasma activated concentrated hydrogen peroxide vapour in sterilizers . In contrast, high level disinfection does not kill endospores but it is used for instruments that don't enter sterile bodily cavity ( eg., Colonoscopy) .

5. SOROLATION is the formation of nearly dormant form of BACTERIA. In the limited number of bacteria, spores are preserve the genetic material of the bacteria when the condition are inhospitable ans lethal for the normal form of the bacteria .

Sporolating bacteria can take in two distinct morphologies , one being an endospore and the other being vegetative cell.

6. a- there will be no moist and temperature will be increases .

b- it would be damaged .

c- there will be some differences occurs in the process.

d- there will be smear of a clostridium species .

f- they may show a mixture on gram positive and negative in a pure culture.
