HIV-1 RT thumb and fingers serve as a clamp aligning template primer of RT correctly toward the polymerase active site.

From the template grip, the active site of p66 contains three catalytic residues which are theorized to aid in the binding of the template primer.This active triad is positioned close to the 3' OH terminus of the DNA primer where DNA polymerization starts.
