9 (B)(1) Aseptic technique is essential so that no contaminating organisms are introduced into culture,to ensure that the contaminating organism does not contaminate the handler and no contamination remains after.
(2) Heat is used to sterilize the mouth of tube before and after obtaining a sample from it in order to keep it from being contaminated.
(3)Air contamination can be prevented when an inoculating loop is used to introduce or take a bacterial sample to/from an agar plate by not opening the lid of the plate fully.
4)The plate should be labelled on the top border of the sampled side of the so that even if the lid is misplaced the plate with the sample is labeled.
5) The agar plates should be incubated upside down inorder to prevent condensation on the agar surface.
6)Disinfectants are effective against vegetative cells and viruses. Disinfectants are not effective aginst endospores. In most of the labs disinfecting bleach wipes are used for this purpose.
