1. The mRNA is transcribed in 5’ to 3’ direction from the template strand (3’ to 5’). The 5’ CAP is a specially altered nucleotide attached to the 5’ end of the mRNA. The 5’ end has a free phosphate as it the first nucleotide incorporated in the chain. An altered GTP is added to the 5’ end by the enzyme guanyl transferase. The 5’ end of mRNA is denoted by G.

2. The poly-A tail is a long chain of adenine nucleotides added to mRNA molecule after transcription. It increases the stability of RNA. It is added to the 3’ end of the mRNA. The 3' end of the transcript is cleaved to free a 3' hydroxyl. To this 3’ hydroxyl end, poly-A polymerase adds a chain of adenine nucleotides. The 3’ end of the mRNA in the figure is denoted by H.

3. The ribosome-binding site (RBS) is present on the mRNA. RBS is a set of nucleotides that are present upstream to the translation start site, where RNA polymerase binds. RBS is required for assembly of ribosomes. RBS is not present in DNA.
