Question

11) Download your lab section's class data from cuLearn. Use the class data to construct a genetic map of the white (w), miniature (m), and yellow (v) mutations. Look up the actual chromosome map of these mutations in Drosophila melanogaster, and compare it to your experimental map. (8 Marks) 12) Use the class data to calculate the coefficient of coincidence (CoC) and interference values for your experiment (its fine if you get a negative inference). Show your work. (3 Marks) 13) Perform a 2 comparing your lab section's data to what would be expected (based on the known map). Like before check page 19 of the lab manual for the format of this test. (6 marks)

Explanation / Answer

We need to get the frequency of recombination of each pair of genes:

FR = RECOMBINED/TOTAL * 100

FR (y-w) = (82+42+39+30)/816 * 100 = 23.65%

FR (y-m) = (106+103+82+39)/816 * 100 = 40.44%

FR (m-w) = (106+103+42+30)/816 * 100 = 34.43%

The lowest the frequency of recombination, the closest the genes are between each other. So the map would be something like:

y--(23.6)--w---(34.45)---m

|------------(40.44)----------| (y-m)

The real map is actually:

y-(1.3)-w---------(37.2)--------m

|-----------------(38.5)-------------| (y-m)

Y and w are the closest, followed by w and m and finally y and m.

So the distant between y and w is smaller and the one between w and m is a lot longer, but the order of the genes in the map is the same.
