Answer-1:
(a) Cloning process is also refered as Somatic Cell Nuclear Transfer. Though there are many differnt steps incolved in the cloning process in order to generate genetic replicas but the main one is called Reproductive cloning technique. Cloning involves the following steps:
1. Nuclear Transfer- The genetic material (DNA) is tranferred from one animal cell to the cell of another animal whole nucleus has been removed in order to be cloned.
2. Stimulation of Cell Division- The transplantation of the egg takes place containing the complete infromation of an organism to be cloned. This implanted egg will act as totipotent which will have an ability to create a whole new organism by cell division. however, cell division is stimulated by treating the egg with certain chemicals or electric shock.
3. Embryo Transplantation in Host- Once the stimulation has been provides, cell divides and forms a multicellular embryo. When the enough development of this embryo has been done, they are taken up and transferred or implanted into the uterus of a female host which will be acting as a surrogate mother; so that the rest of the development of the mebryo could take place inside the womb.
(b) Methods to cmake an animal a trangenic involves:
1. DNA Microinjection- In this, a selected gene is transferred to male pronucleus so that after fertilization, modified fertilized ovum gets transferred to fallopian tubes where implantation occurs and further development could take place.
2. Embryonic stam cell mediated gene transfer- In this method, desired DNA is insertered into in-vitro culture of embryonic stem cells by homologous recombination.
