Individual bacterial cells are too small to see with our naked eyes but they can be observed by powerful microscopes.Eventhough individual bacterial cells are unable to identify with our naked eye they can be observed when they grow as colonies either in liquid medium or solid surface like agar gel.In liquid medium bacteria divide by binary fission until nutrients are available then few drops are placed in petri dish containing agar gel through plating process.Then the cells undergoes binary fission and forms clump of bacterial cells which are immobilised in gel.When the number of these cells reaches 107 then the colony can be observed through our naked eye.
When colonies of bacteria grows on solid culture media they are used to give characteristics of different types of colonies grown on it based on their shapes,size,colour,texture,elevations and margins.They appear in the form of filamentous,circular,rhizoids,irregular,convex,flat,filiform,lobate,curled etc.,same species exhibit same characteristic properties based on that we can identify them from other group of colonies.
