Purpose of all these procedures is that to know about the microorganism s' morphology.
Techniques learned - smear preparation, gram staining, endospore staining, Zeihl-Neelsen stain.
Results can be obtained from the slides observed under the microscope. It helps us to deduce whether the organism is gram positive or gram negative, supports endospore formation, is acid-fast or not. The results gives detailed morphology of the microorganism.
What I learned in this lab is handling of microscope, preparation of smear on slide, about few staining techniques like - gram staining, endospore staining, Zeihl-Neelsen staining. Principle behind different techniques was also understood.
Smear preparation is very important and crucial step in any staining technique. So, it has to made sure that a proper smear is made. Also, staining should be done properly to obtain beautiful stained slide. Exposure to different stains should be properly timed.
