Question
8) (10 pt) A newly discovered antibiotic "macrolide z24" is thought to inhibit one of the electron transport ATP synthase. To determine the site of action of macrolide 724 vou perform the following three experiments Exp. #1 : When excess macrolide Z24 is added to a suspension of respiring mitochondria and P. the concentrations of NADH and FADH, both increase after a few minutes, and O2 consumption and synthesis both decrease. Exp. 2: when an excess of TMPD is aded to the inhibited reaction mixture from Exp., there isno consumption and no ATP is synthesized (TMPD transfers electrons to cytochrome c to produce Exp. 3: When an excess of ferricyanide is added to the inhibited reaction mixture from Exp #1, there consumption and no ATP is synthesized (Ferricyanide removes complexes or ATP is no change in electrons from cytochrome c to produce oxisdiznd (a) Which proteins or protein complexes are Dledot as targets of macrolide n4 by the results of Exp. #1? Explain. (b) which proteins or protein complexes are nic #3? Explain. out as targets of macrolide 224 by the results of Experiments #2 and (c) Which protein or protein complex is most likely the target of inhibition by macrolide 724? (d) If your conclusion re: the site of action of macrolide 724 is correct, what should happen when you add 24- dinitrophenol (DNP) to the inhibited reaction mixture from Exp. #1? Explain.Explanation / Answer
Ans-A- in the first experiment the ATP synthesis is inhibited which indicates that the ATP synthase is ruled out in this experiment.
Ans-B in second and third experiment ATP synthesis is reduced which clearly indicated that here also the ATP synthase activity is being ruled out.
Ans-C here in most experiment the ATP synthase is inhibited by the macrolide Z24. It works as an inhibitor for the ATP synthase which caused the decreased concentration of ATP.
Ans-D- DNP is also an inhibitor just like microlid Z24 so it will also show same results.
