"Abstract

Dental root caries are a common appearance and have been found to be the work of bacteria, pH, and acid levels within the afflicted patients tooth biofilm. A different form of tooth decay may have been identified by having unique conditions. The unique tooth decay is characterized by a single tooth per patient revealing a uniform circular hole in the cementum inside the root. The decay is isolated, with no other teeth being affected by the decay. Additionally, this unique tooth decay is found in both healthy patients and those affected by gingivitis.

To identify the bacteria responsible for the highly specific single tooth decay by utilizing article research of similar instances and affected area bacterial isolation and identification.

The following search strategy to be utilized to identify all possible relevant published articles: First search of Pubmed/Medline and Cochrane Library electronic databases for instances of single root caries; Secondly a search for additional references from the references section of each relevant, electronically-retrieved article. Once all possible relevant published articles are identified. The articles fitting the preceding criteria will then be further screened by researchers searching for identifiable bacteria or mention of isolated uniform root   caries. Analyzation of similarities between instances will be logged and further researched.

Bacterial Isolation and Identification methods: Preliminary research from a small group of dentists has revealed Porphyromonas gingivicanis, Porphyromonas circumdentaria, Pasteurella multocida and Brachymonas chironomi to be the most prevalent bacteria in patients exhibiting the unique tooth decay. To increase the power of the results, Dentists in the Chicagoland area can be asked to report any instances of the unique tooth decay. Swabs may be taken and sent to a single testing area. An increased number of subjects will aid in the identification of the bacteria responsible for the unique tooth decay. The unknown bacteria will be isolated and identified by swabbing the decaying area of the tooth as one sample and collecting a control sample from the same patient from an unaffected tooth. Identification of the bacteria will be done by (in succession) DNA extraction from the samples, 16S amplicon sequencing, High-throughput sequencing on the MinION platform from Oxford Nanopore, Homology searches against the NCBI 16S database, and finally sifting through the results using a custom Perl script.

Predicted Results: Of the four bacteria identified in the preliminary results, two will be dismissed by increasing the population and finding them statistically insignificant in the areas affected by the unique tooth decay."

Explanation / Answer

The identification of bacteria causing the single dental caries is highly needed for treatment purpose. Most of the cases common antibiotics are used to treat without detection the specific bacteria which unable to cure the disease properly and increase the chance of reoccurrence.
