19) (B-gal) How would you make 5 mL of 0.013 M o-nitrophenol-b-D-galactoside (ONPG)?
20) (DNA isolation) How would you make 250 mL of 0. 1 M EDTA solution?
Explanation / Answer
19.
MW of IPTG = 238.3 g/mol
M = 0.013
V = 5 mL
M = (W/MW)(1000/V)
W = (M X MW X V)/1000
= (0.013 X 238.3 X 5)/1000
= 0.01548 g
= 1.5 mg
Weigh 1.5 mg of IPTG and dissolve it in 5 mL of water.
20.
Stock = 0.5 M
Required concentration = 0.1 M
Required volume = 250 mL
C1V1 = C2V2
V1 = (C2V2/C1)
= (0.1 X 250)/0.5
= 50 mL
Take 50 mL of 0.5 M EDTA stoack and add 400 mL of water.
