Question

You are responsible for your assigned microscope! Microscope Storage Proper preparation of the microscope for storage reduces the chances of damage, and ensures that it will be ready for use by the next student. You should know and follow the proper steps for storage. Date Microscope #- Prism sliding bar all the way in? No slide on stage? Stage lowered fully? Mechanical stage retracted? 4X objective returned to viewing position? Rheostat returned to lowest setting? Power switch off? eionfor Futher Though and Revitor Why must you center and focus the object in the field of view under low power before switching to high power ? 1. 2. Why is only the fine adjustment used for high power ? 4. Describe how each of the following is affected by switching from the low to high power objective. total magnification - diameter of field- brightness of field - working distance- depth of focus - Why must a specimen viewed with a compound microscope be very thin? Why are specimens sometimes stained with dyes? 5. 54

Explanation / Answer

1. The 4x objective lens has the lowest power and, therefore the highest field of view. As a result, it is easier to locate the specimen on the slide than if you start with a higher power objective. It is easier to focus in low power than in high power.

2. First of all fine adjustment will help in more detailed and more finer view. To see the internal details of an object fine adjustment is used. If we use coarse adjustment with the high power it will result in crashing the stage.

4 Light intensity, depth of field, working distance, decreases

Magnification increases

5. So that we have a proper view of internal structures. Cells are stained with dyes to differentiate them with the surrounding or to view them better.

7. Nucleus- both- Brain of the cell. it regulates all the activities taking place inside the cell.

Cell- wall- Only plants- helps in protection

Chloroplast and amyloplast in plants- chloroplast contains chlorophyll and helps in photosynthesis whereas amyloplast helps in storage of carbohydrates.

The cell membrane is found in both and it is selectively permeable.

Cytoplasm- Present in both. Here all the metabolic reactions take place.
