After protein enter into the ER lumen N-terminal of protein is cleaved by peptidase present in the ER lumen. N-terminal of protein rich in hydrophobic/ non polar amino acids it makes a transmembrane alpha helix in intermembrane space of ER. The remaining part enter into the ER lumen then peptidase cut the N-terminal. The function of the rpotein is not affect by this process.
