1. You are eluting a fragment of DNA from an agarose gel. You load 5 l of the DNA solution into one well of the gel. When you photograph the gel and measure the fluorescence compared to the standards, the amount of DNA is 50 ng. What is the concentration of DNA in the original tube? You elute the band and rehydrate the final pellet in 10 l of 0.1X TE. You load 1 l of that solution onto another gel, and after electrophoresis, you photograph the gel and estimate that the amount of DNA in that lane is 5 ng. What is the concentration of DNA in the tube containing the eluted tube, and what is the percent of the original DNA that has been recovered after elution?

you take 5 l of DNA solution into one well of a gel and found that he DNA concentration is 50ng, then the concentration of DNA in the original tube is - 10ng/ l.
As you are loading 5 l which gives a band corresponding to 50ng, therefore the concentration is 50/5= 10ng/l
Now, we elute the band and rehydrate in 10 l of 0.1X TE buffer. 1 l from this solution was loaded into a gel. If the DNA concenration is 5ng, then the concentraiton of DNA in the eluted tube is 5ng/l
Now, original DNA was 10ng/l .and the eluted DNA is 5ng/l. Thenrefor 50% of the original DNA has been recovered after elution.
