Question
7. (5 points) GnRH is a hypothalamic neuropeptide that controls mammalian reproduction. Total RNA was extracted from the rat hypothalamus tissue and Northern blot analysis was performed with oligo probes complementary to the 5'end of GnRH mRNA to detect this mRNA. Lane 1 shows blot of control in vitro synthesized GnRH mRNA (560nt). Lane 2 shows blot of the total RNA extract. 4300 nt Explain why lane 2 displays a large smear with several major bands showing above the mRNA band. What is the mechanism that generates these additional bands? mRNAExplanation / Answer
Total RNA contains all types of RNAs. Lower most band is of tRNA and it is usually the brightest one in agarose gel. Next band is of 5S rRNA and then 16S and 23S. rRNAs.Apart from these on the topmost a plasmid or compacted genomic DNA band may occur.
