Question
If I prepared a slide for a Gram stain from a culture of Aeromonas hydrophila and gave it to you to observe under the microscope, what cell shapes and of what color would you observe?
You inoculate 999mL of fresh, sterile media with 1mL of a culture grown overnight to a cell density of 1x106 CFU/mL. What is the cell density (CFU/mL) of the 1L of fresh culture? Please show your work (5pt) and the final answer with the correct units (5pts)

What is the cell density (CFU/mL) of the 1L of fresh culture? Please show your work (5pt) and the final answer with the correct units (5pts).Explanation / Answer
1. Red Rod.
aeromonas hydrophila Gram-negative, straight rods bacterium. Due to gram negative bacteria in gram stain it appears as Red or pink color.
Gram positive looks like Purple.
2. 1ml in 9ml gives 1:10 dilution.
1ml in 99ml gives 1:100 dilution
1ml in 999ml gives 1: 1000 dilution.
So, 1x106 CFU/mL in 1ml mixed with 999ml gives 1:1000 times dilution
1x106 CFU/1000 ml = 1x103 CFU/mL
= 1x103 CFU/mL
