When we add radiolabeled sulfer in the medium so, radiolabelled amino acids are produced. These radiolabelled amino acids are incorporated into the protein of interest and takes place in mitochondria.
When radiolabelled protein of interest is present in the mitochondria then its radioactivity is measured. If it shows radioactivity then we can say protein of interest is present. When radioactivity os not obtained from mitochondria then no protein of interest in mitochondria.
Peroxispomal and other protein does not have role in this determination because we clearly go with mitochondrial analysis.
