The first question can be answered looking at the steps of the sequencing and choosing from there. I will choose two that I like and explain briefly:
1) Ligation of adapters: Once the DNA is fragmented in shorter parts (200-600 base pairs), short sequencies of DNA called adaptors are attached to this fragments in order to allow them to bind to a flow cell and also be amplified by PCR.
2)Adding bases: The first sequencing cycle begins by adding four labeled reversible terminators, primers, and DNA polymerase. Each of the terminator bases (A, C, G and T) give off a different colour.
Next, we can answer the c) question: Lets circle the upper left cluster and follow that one.
The first base added was Tymine (in red). Next: Tymine again. Next: cytosine. Next: cytosine.
Sequence would be: TTCC
If we choose the upper right cluster it would be: CCCG
