Question

Can someone help me with these questions?

Question 13 (15 points) You would like to continue to study the Holt-Oram Syndrome covered in class last Friday. 1.You want to identify all the targets of this transcription factor in the heart and the limb. Using methods from Chapter 3, describe an experiment to do this. State whether your experiment is a Find It, Lose It or Move It experiment? (You have an antibody to the mouse Tbx5 protein if that helps) 2. Your best candidate target of Tbx5 in the limb is a another transcription factor Spalt-like1 (SALL1). Describe an experiment that would provide correlative evidence (Find It) that your candidate is a target of Tbx5 in the limb. 3. We left the discussion open on Friday as to what might be a means to generate a more severe phenotype on the left side in the imbs. We know that the Tbx5 mRNA is expressed at equivalent levels in both left and right limb primordia. Given what you know about the many levels at which a gene's expression can be regulated, propose a testable hypothesis for what could generate the left-sided severity in the limb. (you do not need to describe the actual test) PLEASE keep your answers brief! ab Save

Explanation / Answer

Ans- 1 the method we use will be "find it" . In this we will use flourscene or we will use labelled transcription factor, when it will bind to some other factor then that will be detected by the measuring the fluoroscence of that specific component of heart and limb.

Ans-2 it can be performed by the same method as described in first part, that specific transcription factor can be labelled by using some marker when the target will meet with transcription factor they will produce fluoroscence.

Ans-3 the severity in the limb can be produced by producing some mutation in the target sequence of left limb or by altering the transcription factor receipient in right limb.
